Comparisons of the effects of Bacillus subtilis protease, type VIII (subtilopeptidase A), and insulin on isolated adipose cells. II. Antilipolytic action.

چکیده
The effects of Bacillus subtilis protease, Type VIII (subtilopeptidase A, EC 3.4.4.16), on the lipolytic action of norepinephrine, corticotropin, caifeine, and theophylline on isolated adipose cells were examined. The results indicate that B. subfilis protease, at concentrations suboptimal and optimal for the enhancement of glucose utilization, inhibited, like insulin, the lipolysis mediated by the above agents. Adipose cells, pretreated with insulin or B. subfilis protease, followed by extensive washing, still exhibited elevated glucose utilization and a reduced response to the lipolytic hormones and compounds. The inhibitory action of B. subfilis protease on the stimulated lipolysis was reduced with increasing concentrations of the lipolytic agents. Other proteolytic enzymes such as trypsin, a-chymotrypsin, and Sfrepfomyces griseus protease (Type VI), also exhibited an antilipolytic action. The antilipolytic action of proteolytic enzymes, which was found to be independent of their ability to stimulate glucose uptake, is considered to result from the proteolytic modification of the plasma membrane of the adipose cells via as yet unknown mechanisms.

The goal of this research was to isolate and identify the thermostable alkaline protease producing bacteria among several native Iranian microorganisms. At the end of screening program, a Bacillus subtilis BP-36 strain producing thermophilic alkaline protease was isolated from a hot spring in Ardebil province. The target enzyme was purified using a one-step Aqueous two-phase systems (ATPS) prot...

Objective(s): We aimed to characterize the phenotype and genotype of Bacillus spp isolated from diabetic patients’ eyes, by studying the drug sensitivity patterns with a disc-diffusion method. Materials and Methods: Fifty eyes of 25 patients with type II diabetes mellitus, with at least 10 years of diabetes history, were included in the study. We analyzed the eyes for the presence of Bacillus ...

The present study explored the efficacy of Bacillus spp. and protease production for biocontrol of the root-knot nematode Meloidogyne javanica in African violet media. Among 100 bacterial isolates from various soils, the highest nematode mortality was observed for treatments with isolate GM-18, which was identified as Bacillus subtilis based on cultural and morphological characteristics and 16S...
متن کاملSimultaneous Effects of Metformin and Sitagliptin on the Contents of Insulin Resistance Proteins Glucose Transporter 4 and Protein Kinase B in Diabetic Patients\' Adipose Tissue
Objective: Obesity is a factor in the development of insulin resistance and type 2 diabetes. Obesity contributes a wide variety of metabolic changes such as insulin resistance. The insulin signal mechanism to intra-cells occurs in insulin resistance, primarily in adipose tissue cells, which can be appropriate targets for therapeutic approaches by recognizing the proteins in this pathway. The st...
